Capacitation is a hormone dependent event in the spermatozoa, as they enter the female genital tract. Capacitation must occur before a spermatozoon can fertilize a mature oocyte and it is considered as the ultimate event in the maturation process of spermatozoa. Capacitation results in destabilization of sperm membrane and increasing permeability leading to acrosome reaction. The sperm cannot get back its original form once acrosome reaction has occurred. But capacitation is reversible. Capacitation is promoted by the process of separation of sperm from seminal plasma. This occurs during the passage of the sperm through the cervical canal, during natural insemination. Seminal plasma contains several de-capacitation factors. Is the laboratory practicing IUI or IVF, separation of plasma from the spermatozoa is one of the important procedures to produce capacitation
